Clinical Definition

Granulomatosis with Polyangiitis is a systemic vasculitis characterized by necrotizing granulomatous inflammation and small to medium vessel vasculitis. It is an autoimmune disorder typically associated with the presence of anti-neutrophil cytoplasmic antibodies (ANCA), especially c--ANCA directed against proteinase 3 (PR3). The disease primarily affects the upper and lower respiratory tracts and kidneys, leading to granuloma formation and glomerulonephritis. Clinical significance lies in its potential to cause life-threatening organ damage if untreated. The pathogenesis involves immune-mediated endothelial injury and granulomatous inflammation.
